Topics:            Two major protein degradation pathways exist in eukaryotic cells: autophagy and the ubiquitin proteasome system (UPS). Whereas the UPS is responsible for the rapid degradation of proteins when fast adaptation is needed, autophagy is mainly involved in the degradation of long-lived proteins and entire organelles. Autophagy plays an important role during development, tumor suppression, immunity, tissue homeostasis and is required for the adaptation to environmental stresses such as starvation.

The aim of this international conference series is to bring together scientists working on autophagy and ubiquitin-dependent mechanisms. The second medical conference of this series will be devoted mainly to mechanism and regulation of autophagy with a particular focus on ubiquitin mediated selective autophagy, different aspects of autophagosomes biogenesis, and the relationship between autophagy and apoptosis. We will also cover different physiological and pathological aspects of this process with a special emphasis on cell death, immunology, neurodegeneration and cancer.

Topics:            The organized trafficking of leukocyte subsets through the tissue compartments of the body is critical to the generation of innate and adaptive immune responses and for normal organ immuno-surveillance; however, when misguided, such processes can promote uncontrolled inflammation resulting in tissue pathology. It is now well established that cellular adhesion and chemokine receptors play a central role in regulating tissue-specific leukocyte migration. Recent studies have started to unravel the complex mechanisms underlying the expression of these receptors on lymphocytes and highlight the importance of the local lymph node environments and migratory dendritic cells in the induction of peripheral tissue homing receptors. While immunologists have traditionally tended to focus on primary and secondary lymphoid compartments to study immunological processes and immune cell interactions, there is a growing realization that many, if not most, of our immune cells are located in peripheral tissues and that each of these sites possess unique immunological characteristics. Without mastering the large-scale molecular data that underlies pathophysiological states, without sophisticated mathematical algorithms capable of data integration, and without an appropriate informatics infrastructure to apply these algorithms and translate the results into manageable bites of information that can be consumed by physicians and patients, efforts to realize the dream of personalized medicine will be difficult to achieve. Topics:            Apolipoprotein E4 (ApoE4) genotype is the primary risk factor for late-onset Alzheimer?s Disease. Approximately one fifth of the human population are ApoE4 carriers, yet more than 17 years after its discovery, the molecular basis by which this common allele causes neurodegeneration is still not understood. One major reason is the lack of effective communication between lipid researchers who have investigated the role of ApoE as a plasma lipid transport protein, and neuroscientists who are unfamiliar with the complex biochemistry of lipid metabolism and the cell biology of ApoE receptors. This is confounded by the methodological difficulty of studying lipid metabolism and lipoprotein transport in the brain and the paucity of pioneering work tackling this problem. Yet, the solution to one of the most pressing socioeconomic problem of our times, the looming Alzheimer?s Disease epidemic, depends on our ability to dissect and understand the molecular mechanism by which ApoE4 is causing this disease. The purpose of this highly interdisciplinary medical conference on ApoE, Alzheimer?s and lipoprotein biology is to bring together a critical mass of experts with complementary expertise from these different fields, but who do not normally collaborate or attend the same conferences. In this manner, we envisage this medical conference serving as the main catalyst for communication and collaboration between lipid and lipoprotein researchers, neurogeneticists, cell biologists and synaptic neurophysiologists. Opportunities for interdisciplinary interactions will be significantly enhanced by the concurrent medical conference on Clinical and Molecular Biology of Acute and Chronic Traumatic Encephalopathies, which will share opening keynote addresses and a fourth day of sessions with this medical conference.

Topics:            Acute traumatic brain injury (TBI) and its long-term neurodegenerative complications (known collectively as chronic traumatic encephalopathy, or CTE) are increasingly recognized as major public health issues, especially in the context of sports-related head injury and battlefield blast (military) exposure. Awareness of these issues has risen as neurological, psychiatric and neurodegenerative phenomena are associated with professional athletes and with veterans returning from Iraqi and Afghan theaters. The physics of head injury, as well as the epidemiological and neuropsychiatric aspects of sports-related and blast-related syndromes, will be reviewed in the initial sessions of this three-day medical conference. Advances in functional neuroimaging and in the rapid and convenient determination of peptides and proteins in the cerebrospinal fluid and plasma will be reviewed in terms of their possible pathobiological significance as well as their identities as possible biomarkers. Later sessions will focus on the overlap between CTE and major neurodegenerative diseases, such as Alzheimer?s Disease (AD), frontotemporal dementia and amyotrophic lateral sclerosis. CTE is associated with accumulation of aggregated proteins or protein fragments, including tau, TDP-43, the Alzheimer?s amyloid precursor protein (APP) and the APP metabolite, amyloid-b. New experimental therapeutics research targets the roles these abnormal protein structures play in injury severity and subsequent outcome. Ab-lowering medications, currently in clinical trials for AD, improve outcome following acute brain trauma in rodent models, suggesting a pathway toward potential therapeutic interventions. Because apolipoprotein E (APOE) isotype is an important determinant of outcome from acute severe as well as mild repetitive TBI, this medical conference coincides in time and venue with the Keystone Symposia medical conference on ApoE, Alzheimer?s and Lipoprotein Biology. Opening plenary sessions and an optional fourth day of sessions are available for those interested in learning more about basic biology and/or clinical interventions related to APOE.

Topics:            In recent years there have been major advances in our understanding of the mechanisms that are involved in the recognition of microbes and subsequent activation of host immune defenses. Several classes of membrane-bound and cytosolic pattern recognition receptors (PRRs) have been identified and partially characterized. These include the Toll-like receptors (TLRs), NOD-like receptors (NLRs) and RIG-I-like receptors (RLRs). Activation of PRRs leads to the production of a large array of pro-inflammatory and anti-microbial molecules that are critical for the elimination of invading pathogens and activation of adaptive immune responses. A class of cytosolic PRRs is involved in the assembly of the inflammasome, a molecular platform that mediates activation of caspase-1 and secretion of mature IL-1b and IL-18. Importantly, activation of the inflammasome is also induced by non-microbial mechanisms including endogenous molecules involved in the pathogenesis of metabolic and inflammatory diseases. In addition, there is evidence that autophagy, a cellular process that mediates recycling of intracellular components, is involved in microbial recognition and plays a key role in host defense. The initial sensing of microbes often occurs at mucosal surfaces, but the interplay between recognition of commensal vs. pathogenic microbes on the host immune system is only beginning to be uncovered. The goal of the Keystone Symposia medical conference on Innate Immunity: Sensing the Microbes and Damage Signals is to gather scientists working on innate immunity to discuss cutting-edge research on the mechanisms that regulate the activation of the immune system by microbes as well as by endogenous damage signals, and to integrate such knowledge in the context of inflammation, homeostasis, host defense and disease. The integration of these divergent disciplines has allowed us to develop a clearer definition of a functional b cell and has elucidated many of the mechanisms underlying the differentiation, survival and death of the b cell. Furthermore, these studies have revealed the potential to generate new b cells through replication or neogenesis. Improvements in differentiation and transdifferentiation protocols have also illustrated the potential of using stem cells and some somatic cell types as sources of islet cells, but these studies have also revealed the challenges associated with generating functional b cells. The purpose of this Keystone Symposia medical conference is to highlight the areas of islet biology that have seen the most remarkable advances and to address some of the controversies that still exist in the field, especially with regards to the generation and maintenance of functionally mature b cells for therapeutic purposes. Breakthroughs in bladder and tracheal reconstruction and highly promising approaches for solid organ regeneration as well as musculoskeletal and nerve repair have been achieved. Effective cell sourcing, optimal assembly and fabrication of tissue-derived and synthetic biomaterial scaffolds, and targeted regulation of therapeutic and immunomodulatory functions of stem cells represent some of the current challenges in this field.
